"Desperado" as performed by Linda Ronstadt brings a lump to my throat every time. Which is kinda strange, as generally I don't care for country/western at all.

I think that for the most part, the memory linked to the music is what affects me, rather than the song itself. I have hundreds of songs, each that remind me of certain things, almost always people I've loved and lost for one reason or another. Those are the one's that have power over me.
The exception to that is The Show Must Go On by Queen. I find it absolutely impossible to hear that song without seeing in my minds eye the video where Freddie looks pretty much ready to collapse.

"Texas Heat" from The Beat Farmers sends shivers down my spine everytime I hear it. This is mainly because of the low, emotional voice of Country Dick Montana.
"Highwayman" from The Highwaymen and "Hurt" from Johnny Cash have the same effect.

'Forbidden Colours' by David Sylvian & Ryuichi Sakamoto Ooh. That is a good one. Amazing film too. I think at a pinch I'd go for the version without David Sylvian, but both are ace. And British Sea Power's 'Carrion' always gets me in an odd manner as well.

'Hurt' by Johnny Cash. Actually, his version of 'Bridge Over Troubled Water' from the same album is amazing too. 'Fairytale of New York' by The Pogues always gives me a lump in me throat, too. I think that's mainly because it's such a sweet melody.. but it also has fantastic bitter-sweet lyrics. 'Forever Autumn' also gets me every time.
